01

What is FOXO4-DRI?

Plain language first. The technical label stays, but it never stands alone.

Overview

It is a senolytic. That means it is designed to make worn-out zombie cells destroy themselves, so healthy tissue can take over. Animal results were striking. There is essentially no human safety data.

The technical version

FOXO4-DRI is a specially built (D-retro-inverso) peptide that pries apart a partnership between the proteins FOXO4 and p53 inside aged cells.

What it's used for

An experimental peptide designed to clear out zombie aging cells for anti-aging, with striking animal results.

02

How does FOXO4-DRI work?

What it does in the body, and where.

FOXO4-DRI is built backwards on purpose. D-retro-inverso means the amino acids are mirror images assembled in reverse order, which makes it resistant to being broken down.

  1. 01
    It separates FOXO4 from p53
In senescent cells, that pairing is what keeps the cell alive when it should have died.
  2. 02
    The cell then self-destructs
Which is what senolytic means: clearing worn-out cells so healthy tissue can take over.
  3. 03
Old mice recovered fur, kidney function, and stamina
The result that made this famous.
  4. 04
It has never been given to a human
And some widely circulated lifespan-extension percentages attributed to it are simply fabricated.

What else is it used for?

Each use below carries its own evidence level. Approved means regulators have cleared it for that purpose. Investigational means it is in trials. Early research means it is being studied and is not established.

How much, and do you cycle it?

The range, and whether you take breaks.

Research only, cycled. Commonly cycledCommunity practice

Cycled by convention rather than to a tested schedule. No published study establishes a block length or a break length for this one.

How long does it stay in the body?

The half-life, and where the figure comes from.

Nothing published. The 2017 paper that made it well known measured what it did to cells, not how long it lasted.

How do I dose and price it?

Enter your vial and your dose. Pick a mix. We show the draw and the cost.

The vial holds a fixed amount of drug. Adding more water does not make more drug, it just spreads it thinner, so you draw a bigger number on the syringe for the same dose.

07

How do you store it?

Before mixing, and after.

Dry powder in the freezer or fridge. Once mixed, fridge, and use within about a month.

Unmixed lyophilized FOXO4-DRI keeps for a long time cold. Once you add water the clock starts: most reconstituted vials hold up for roughly 28 to 30 days at fridge temperature. Keep it out of light, and do not freeze it after mixing.

08

What are the side effects and cautions?

Known cautions and warnings for this compound.

Who should avoid it

Check these before anything else.
  • Any history of cancer
  • Pregnancy
  • There is essentially no human safety data, so nobody can tell you who should avoid it

Stop and get help

These are emergencies. Seek care.
  • Any severe or unexpected symptom, given the complete absence of human safety data
  • Signs of an allergic reaction such as face or throat swelling, hives, or trouble breathing

Common effects

Expected, and usually mild.
  • Unknown in humans. Animal work showed temporary tissue effects, but there is no reliable human profile

Where this page says nothing is established, that means nobody has studied it, not that a compound is safe.

09

Is FOXO4-DRI approved?

And where the numbers on this page come from.

No. FOXO4-DRI is not approved for any use, anywhere. It is sold labelled for laboratory research only, and there is no legal route to buy it for human use.

What has happened recently

No human safety data exists. The striking results came from mouse studies, and nobody has established whether the same approach is safe in people.

Sources

  • Targeted apoptosis of senescent cells restores tissue homeostasis in response to chemotoxicity and aging, Cell (open repository copy)(2017)
The paper that created the peptide and the only reason anyone has heard of it. FOXO4-DRI was designed to break the FOXO4 and p53 interaction so that senescent cells undergo apoptosis. Everything measured is cells and mice: it blunted doxorubicin toxicity, and it restored fitness, fur density and kidney function in fast-aging and naturally aged mice. The authors' own phrasing is that this held under conditions where the peptide was well tolerated in vivo, which is a dose-finding statement rather than a safety clearance. There is no human study of FOXO4-DRI, and this paper does not claim one.
