01

What is Enclomiphene?

Plain language first. The technical label stays, but it never stands alone.

Overview

It stops the brain from seeing estrogen, so the brain tells the testicles to make more testosterone.

The technical version

Enclomiphene is the trans-isomer of clomiphene, a selective estrogen receptor modulator that blocks estrogen feedback at the pituitary so LH, FSH and testosterone all rise.

What it's used for

A daily capsule taken to raise testosterone without switching off sperm production the way a testosterone gel does.

02

How does Enclomiphene work?

What it does in the body, and where.

Enclomiphene works on the brain rather than on the testicles.

  1. 01
It blocks estrogen receptors in the pituitary
Estrogen normally tells the pituitary that hormone levels are fine. Blocking that signal removes the brake.
  2. 02
    LH and FSH rise
These are the two signals the pituitary sends to the testicles.
  3. 03
The testicles make more testosterone, and keep working
This is the difference people are buying. A testosterone gel replaces the hormone and shuts the axis down. This raises the hormone by driving the axis harder.
  4. 04
    Estradiol rises as well
More testosterone means more raw material for aromatase. People who take it expecting an estrogen blocker are often surprised by their bloodwork.

What else is it used for?

Each use below carries its own evidence level. Approved means regulators have cleared it for that purpose. Investigational means it is in trials. Early research means it is being studied and is not established.

04

How much, and do you cycle it?

The range, and whether you take breaks.

12.5 up to 25 mg per day, daily. Continuous in the trials Trial dose

Both phase 3 trials dosed daily for three months with no planned break, and an open-label extension continued daily 12.5 mg for up to a year. The post-cycle use it is sold for, a short course to restart the axis, is not the way it was studied.

05

How long does it stay in the body?

The half-life, and where the figure comes from.

About 10 hoursHuman study

This is the short-acting half of clomiphene, isolated. Peak concentration comes 2 to 3 hours after a dose. The hormone effect lasts far longer than the drug, with raised testosterone, LH and FSH persisting at least 7 days after stopping.

06

What does it cost?

Typical dose and cost, in plain numbers.

Enclomiphene is not injected

It is taken by mouth (oral). Both phase 3 trials tested exactly two doses, 12.5 mg and 25 mg once daily, for three months. There is no approved label, so there is no official dose above or below that.

07

How do you store it?

Before mixing, and after.

Room temperature. Cool, dry, out of light.

Enclomiphene is taken as a pill or capsule. Keep it in its original container at room temperature, away from heat, humidity, and direct sun. No fridge or mixing needed. Check the expiration date on the bottle.

08

What are the side effects and cautions?

Known cautions and warnings for this compound.

Who should avoid it

Check these before anything else.
  • A history of blood clots, stroke or clotting disorders, which applies to this whole drug class
  • Existing liver disease
  • Any hormone-sensitive cancer
  • Pregnancy, and anyone who could become pregnant
  • Banned in drug-tested sport

Stop and get help

These are emergencies. Seek care.
  • Blurred vision, flashes, floaters or any change in how you see, which is a known class effect
  • Swelling, pain or warmth in one leg, or sudden shortness of breath
  • Yellowing of the skin or eyes, or dark urine
  • Signs of an allergic reaction such as face or throat swelling, hives, or trouble breathing

Common effects

Expected, and usually mild.
  • Headache
  • Nausea
  • Dizziness
  • Mood changes
  • Rising estradiol, since more testosterone means more substrate for aromatase

Where this page says nothing is established, that means nobody has studied it, not that a compound is safe.

09

Is Enclomiphene approved?

And where the numbers on this page come from.

Not approved, but legal to obtain. Enclomiphene has no FDA approval, and it can be prescribed or prepared by a compounding pharmacy in the United States.

What has happened recently

Never approved anywhere. Repros Therapeutics applied for approval as Androxal and received a Complete Response Letter on 1 December 2015, in which the FDA said the phase 3 design was no longer adequate to demonstrate clinical benefit and asked for at least one further phase 3 study. That study was never run and development was dropped. In June 2022 the FDA's Pharmacy Compounding Advisory Committee voted against putting enclomiphene citrate on the final 503A bulks list. It nonetheless still sits in category 1 of the FDA's nominated bulk substances list as of May 2026, meaning under evaluation, and that is what keeps compounding-pharmacy supply legal for now. Prohibited in sport at all times by WADA under section S4.

What regulators say

From official documents

Banned in drug-tested sport. Most research peptides are prohibited under WADA's S0 category, which covers any substance with no approval from any regulatory authority for human use. That means a compound can be banned without ever being named on the prohibited list. Check with your own anti-doping authority rather than assuming absence from the list means permitted.
